You sleep seven or eight hours straight. In that window you eat nothing, drink nothing, and your body gets no fresh supply of amino acids at all. For your muscle it is the longest fast of the day, and it lands in exactly the phase where repair and growth are meant to happen.
This is where a popular idea comes in: a protein shake right before bed. Sounds logical. But is it really worth it, do you need casein specifically, and does it work with plant-based protein too?
The research on this is surprisingly concrete. And it quietly clears up a stubborn myth along the way. What happens in the muscle overnight, and what you can take from it for your own routine: that is what we are getting into.

What happens in the muscle overnight
While you sleep, muscle protein synthesis runs on low. The reason is simple: without a supply of amino acids, your body lacks the building material to make new muscle protein. A review sums it up by noting that the overnight synthesis rate is limited by the available amount of amino acids, and that gap is exactly what a protein meal before bed can close.1
Protein taken before sleep is effectively digested and absorbed overnight and thereby raises muscle protein synthesis during sleep. A robust increase needs at least around 40 g, and prior exercise improves how well the ingested amino acids are used (Trommelen 2016, review).1
For context: this is not an emergency. If you eat enough protein across several meals during the day, you have already done most of it right. The bedtime shake is an extra lever, not a duty. It gets interesting mainly when your needs are high or your evening is otherwise low in protein.
Why casein is called the „night protein"
Casein is seen as the ideal evening protein because it digests slowly. It clots in the stomach and releases its amino acids into the blood over hours rather than in a short spike. Back in 1997 a tracer study already showed this prolonged, moderate amino acid plateau for casein, against the rapid rise after whey (Boirie 1997).2
This split into „slow" and „fast" proteins shaped a concept that still informs every protein recommendation today. The numbers behind it are worth a look.
Boirie and colleagues (Proc Natl Acad Sci USA) used labelled amino acids to track what happens after a single meal. Whey produced a short, high rise in blood amino acids, casein a prolonged, moderate plateau. Casein suppressed protein breakdown by 34 per cent, whey stimulated synthesis more strongly (plus 68 versus plus 31 per cent). Two routes, one goal: keep more protein in the body.2
How fast a protein digests shapes the response after a meal: casein slows gastric emptying and creates a moderate, sustained amino acid plateau that dampens protein breakdown, while rapidly digested whey drives synthesis more strongly (Boirie 1997, tracer study).2
From this mechanism came the rule of thumb „casein for the night". It is plausible, which is exactly why it sticks. Whether that sustained amino acid plateau actually delivers more muscle overnight than a fast source is a separate question, and only newer studies answer it.
Does protein before sleep really build more muscle?
In the short term the answer is a clear yes: protein before sleep measurably raises overnight muscle protein synthesis. A systematic review of nine studies shows that 20 to 40 g of casein about 30 minutes before bed increase whole-body protein synthesis overnight.3
Reis and colleagues (J Sci Med Sport) reviewed nine studies on protein intake before sleep. The result: 20 to 40 g of casein roughly 30 minutes before bed raise the overnight synthesis rate in younger and older men. Over 10 to 12 weeks of resistance training, young men additionally gained muscle mass and strength, while in older men the evidence for that long-term effect was too limited to conclude.3
Here an honest distinction matters, one that often gets lost. The acute rise in muscle protein synthesis is well established. Whether that turns into measurably more muscle mass over weeks shows up mainly in young, training men, not automatically in every group. A higher synthesis rate for one night is simply not the same as a bigger biceps after three months.
Taking 20 to 40 g of casein about 30 minutes before sleep raises the overnight protein synthesis rate in healthy young adults. Possible gains in muscle mass and strength appear after prolonged resistance training in young men, while the evidence in older adults remains limited (Reis 2020, systematic review).3
Is slow really better? The myth check
If the slow amino acid plateau were really the decisive advantage, casein would have to beat fast whey overnight. A controlled study compared exactly that for the first time in 2023 and found no meaningful difference between the two (Trommelen 2023).4
Trommelen and colleagues (Sports Med) gave 36 young men either 45 g of casein, 45 g of whey or a placebo 30 minutes before sleep, after an evening workout. Overnight muscle protein synthesis was measured over 7.5 hours. Both protein sources sat above placebo, but between casein and whey there was no statistically meaningful difference (myofibrillar p = 0.44, mitochondrial p = 0.69). So the slow digestion gave casein no overnight edge.4
The intervals for casein and whey overlap almost completely. Whey even edged ahead numerically, which is no surprise given its higher leucine content. The message holds: it is not the speed of digestion that decides, but that enough high-quality protein arrives in a sufficient amount at all.
This pattern is bigger than the one overnight study. A meta-analysis on protein timing around training found the same thing: once you account for total daily protein, the supposed timing advantage all but disappears.
A meta-regression of 23 studies on protein timing concluded that the decisive factor for muscle gains is total protein intake, not the precise timing around a training session (Schoenfeld 2013, meta-analysis).5
To be fair: that meta-analysis looked at timing around training, not at sleep specifically. So it does not directly prove anything about the bedtime shake. But it supports the same principle the overnight study shows, and that makes the overall picture fairly consistent: amount beats timing.
Does it work plant-based?
The honest answer first: there is no direct overnight study on plant protein yet, the research on night protein uses almost only casein. The answer can still be inferred, because what matters according to the position stand is the dose and the leucine content, not the protein's origin (Jäger 2017).6
From the casein studies one principle carries over: if enough high-quality protein arrives in a sufficient dose, the source is secondary. For plant protein the question therefore shifts away from digestion and towards the amino acid profile.
An effective protein dose should contain roughly 700 to 3,000 mg of leucine plus a full spectrum of essential amino acids, and plant sources should be dosed or supplemented accordingly to reach that threshold (Jäger 2017, ISSN position stand).6
The weak point of many plant sources is the leucine content: per gram of protein they deliver, on average, a little less leucine than whey. The fix is not a return to animal protein but a sufficient dose and an eye on the amino acid profile. The effect grows stronger if you train in the evening. A study in older men shows that an evening training stimulus markedly raises how well the protein is used overnight. So if you hit the gym in the late afternoon or evening, you get more out of the bedtime shake.
Holwerda and colleagues (J Nutr) gave older men 40 g of casein before sleep, with or without evening activity. With prior training, overnight myofibrillar protein synthesis was 31 per cent higher, and 54 per cent of the ingested amino acids reappeared in the bloodstream. So the training itself amplified the effect of the night protein.7
An evening training stimulus raises how well protein taken before sleep is used: in older men, overnight myofibrillar protein synthesis was 31 per cent higher with prior training than without (Holwerda 2016, RCT).7
Who it is worth it for: anyone who trains in the evening, has elevated protein needs, or otherwise misses their daily target. If you already eat plenty of protein spread across the day, you do not strictly need it. It is an optimisation, not a must.

But the much-cited casein advantage falls apart on a closer look: head to head, slow casein was no better than fast whey. What counts is the dose and leucine, not the speed of digestion.
